The PCR buffer without MgCl2 in combination with the MgCl2 stock SOLUTIONon is used for the individual adjustment of the Mg2 concentration in the PCR reaction. In most applications a concentration of 1.5mM MgCl2 will yield satisfactory results with a dNTP concentration of 200muM each. In many cases however it is required to titrate the optimal Mg2 concentration to increase the specificity and yield of the PCR reaction.

Hot-Start Taq Mastermix™ is the heat activated version of Denville's 2X, ready to go reaction mixture. It is inactive at room temperature and only requires addition of template, primers, and water. A 5 minute pre-heat step at 95ÝC is required to successfully carry out primer extensions. The Blue version contains a non-toxic, non-hazardous inert blue dye, which allows users to ensure adequate mixing and acts as a loading buffer following completion of the PCR reaction. Hot-Start Taq Blue DNA Polymerase is supplied with an inert blue dye that allows the end user a method to confirm addition of the enzyme to the reaction. The dye further allows the user to directly add a post-reaction sample directly into an agarose gel, eliminating the need for a separate gel-loading buffer.
